摘要 目的:研究脾阳虚大鼠脾失健运相关蛋白mRNA表达差异。方法:利用现代分子生物学RT-PCR的实验方法,运用蛋白组学技术鉴定大鼠回肠组织中与脾阳虚相关的2个蛋白,即结蛋白(Desmin)、甘油醛三磷酸脱氢酶(glycer-aldehyde-3 phosphate dehydrogenase,GAPDH),从基因水平检测蛋白质mRNA的差异性表达。结果:Desmin的mRNA表达在正常组和脾阳虚组没有明显差异,GAPDH的mRNA表达在正常组和脾阳虚组有明显差异。结论:GAPDH可作为脾阳虚证的辅助检测指标之一。用分子生物学RT-PCR检测方法检验了两个蛋白mRNA在转录过程中是否发生变化,从结果中看到了蛋白组学筛选的GAPDH在转录过程中发生变化,充分证明了GAPDH与脾阳虚证有关。Desmin的mRNA表达在脾阳虚大鼠中与正常组没有明显差异,还需要进一步进行验证。 Objective:To research the spleen failing to transport and transfom in spleen - Yang deficiency rats and health games related protein mRNA expression differences. Methods : The modem molecular biology of RT - PCR method was used, and application of Proteomics technology was for identification in ileal tissue of rats with spleen-Yang deficiency two proteins : Desmin and GAPDH, detection of mRNA differential expression of proteins from gene was also made. Results : Desmin mRNA expression in normal group and the group had no significant differences due to spleen-Yang deficiency ,while GAPDH mRNA expression in nor- mal and spleen-Yang deficiency of groups had distinct differences. Conclusion : GAPDH can be used as one of the secondary index of spleen-Yang deficiency. Using RT - PCR for detection of molecular biology methods verifies that the two proteins mRNA in transcription is changed in the process. GAPDH Proteomics filter changes in the transcription process, proving the GAPDH is re- lated with spleen-Yang deficiency syndrome. Desmin mRNA expression in spleen - Yang deficiency rats and normal groups do not have obvious differences ,which also requires further validation.
