“脾为气血生化之源”生血、统血机制的实验研究 被引量:4

摘要 目的:以造血干细胞因子对红系造血系统调节的角度,探讨"脾胃气血生化之源"生血、统血的作用及机制。方法:采用经典的复合因素造模法建立脾气虚证小鼠模型和采用苯皮下注射造模法建立再障贫血模型,运用血细胞计数板镜下计数外周全血细胞数,运用直接免疫荧光标记法检测骨髓CD34+及ELISA法检测血清SCF含量。结果:3组相比较,两模型组小鼠外周血象均明显下降,骨髓CD34+与血清SCF的含量也明显降低;脾气虚证模型组与再障贫血组相比较,再障贫血组小鼠骨髓CD34+与血清SCF的含量的降低程度低于脾气虚证模型组小鼠。结论:脾气虚证小鼠骨髓CD34+与血清SCF的含量明显降低,为脾为气血生化之源提供了现代生物学依据。 Objective : From the aspect of hematopoietic stem cell factor acting on erythroid hematopoietic system, to explore the role and mechanism of the spleen being source of qi and blood. Methods : Composite method was used to establish the spleen qi de- ficiency model and the aplastic anemia model was built by injecting benzene in subcutaneous tissue. The hematocytometer was used to count the whole blood cell count ; CD34 ~ in bone marrow was detected by direct immunofluorescence as biomarkers. The contents of SCF in serum were detected by ELISA. Results : Two model mice peripheral blood was significantly decreased. Com- pared with the control groups, CD34~ in bone marrow and content of SCF in serum reduced obviously in the spleen qi deficiency model. Compared with the spleen qi deficiency mice, CD34 ~ in bone marrow and content of SCF in serum in the aplastic anemia mice were lower than. Conclusion:CD34 + in bone marrow and content of SCF in serum of the spleen qi deficiency syndrome in mice were decreased obviously, providing science evidence for the research of spleen being source of qi and blood.
