摘要
目的:探讨巨噬细胞集落刺激因子(Macrophagecolony-stimulatingfactor,M-CSF)抗单核/巨噬细胞氧化损伤的作用机理,揭示M-CSF对小鼠巨噬细胞系RAW264.7细胞抗氧化系统影响。方法:以L929细胞条件培养液为M-CSF来源,采用酶活性测定等生化测定方法,考察M-CSF细胞内还原型谷胱甘肽含量、脂质过氧化物含量、过氧化氢酶活性、谷胜甘肽还原酶活性等检测指标的影响。结果:M-CSF处理可提高RAW264.7细胞内的过氧化氢酶活性及还原型谷胱甘肽含量,降低脂质过氧化物含量,且使细胞在tbOOH攻击下保持较高的抗氧化水平。结论:M-CSF可能通过改善单核巨噬细胞的氧化还原代谢而减轻细胞的过氧化损伤。
objective: In order to find out the mechanisms that macrophage colony-stimulating factor (M-CSF) exerted its functions in alleviating the tert-butyl hydroperoxide (tbOOH)-induced oxidative injury to monocytes/macrophages. Methods: M-CSF riched L929 cell conditioned, medium (L929-CM) was used as the source of M-CSF.The antioxidant system in RAW264.7 cells were investigated by applying biochemical analysis such as enzyme activity detection. Results: M-CSF was showed to improve catalase activity and the content of reduced glutathione, while decrease the content of lipoperoxide in RAW264. 7 cells. M-CSF-treated cells also maintained high antioxidant capability when tbOOH attacked. Conclusion: M-CSF could. alleviate the oxidative injury to RAW264 .7 cells through improving their redox status.
