柿叶黄酮对阿霉素肾病大鼠肾组织TIMP-1表达的影响

摘要 目的观察柿叶黄酮(PLF)对阿霉素肾病大鼠肾组织金属蛋白酶抑制物1(TIMP-1)表达及对肾组织病理损伤的影响。方法制备阿霉素肾病大鼠模型及药物干预模型鼠,测定模型鼠24 h尿蛋白(24hUP)变化,用免疫组化法检测肾组织中TIMP-1表达,光学显微镜观察肾组织病理形态。结果 PLF能明显降低阿霉素肾病大鼠24hUP(P<0.01),减轻肾脏免疫病理损伤及抑制肾组织TIMP-1的表达(P<0.01)。结论 PLF能降低模型鼠肾组织TIMP-1的表达、抑制肾组织纤维化等,对阿霉素肾病大鼠肾脏起到保护作用。 Objective To observe the effect of persimmon leaf flavonid(PLF) on the expression of TIMP-1 in kidney of adriamycin nephropathy(AN) rat.Methods AN rat model and drug intervention rat model were prepared.The expression of TIMP-1 was detected with immunohistochemical method.The pathomorphological change of kidneys was detected by microscope.24-h urine protein(24hUP) was detected in different times.Results PLF could obviously reduce 24hUP,improve pathologic damage of kidneys and inhibit the expression of TIMP-1 in kidneys of AN rats(P〈0.01).Conclusion PLF can protect the kidneys of AN rats by down regulating expression of TIMP-1 and inhibiting tissue fibrosis,etc.
