表皮生长因子对大肠癌细胞游离Ca^(2+)和环磷酸酰苷的影响

Effects of epidermal growth factor on free calciumion and adenosine cyclic phosphate in colorectal carcinoma cells

摘要 目的 探讨表皮生长因子 (EGF)对大肠癌LoVo细胞内Ca2 + 和环磷酸酰苷 (cAMP)的影响。方法 采用激光扫描共聚焦显微镜图像分析技术测定EGF对LoVo细胞Ca2 + 的影响 ;放射免疫技术测定EGF对LoVo细胞内cAMP的影响。结果 EGF能非剂量依赖性引起LoVo细胞[Ca2 + ]i升高 ,40~ 6 0s达到峰值 ,平均升高分别为 (143± 14) %、(147± 15 ) %、(146± 11) % ;加入EGF后 ,LoVo细胞内cAMP快速下降 ,10~ 2 0min降至最低值 ,然后缓慢回升 ,约 90min恢复至基础水平 ,cAMP基础水平平均值为 (48.0 2± 4.6 1)× 10 -8mol/L。结论 EGF可引起LoVo细胞内Ca2 +和cAMP代谢失调。 Objective\ To investigate the effects of epidermal growth factor (EGF) on free calcium ion and cAMP in LoVo cells of colorectal carcinoma. Methods\ Laser scanning confocal microscopic imaging analysis technique was used to investigate the effect of EGF on Ca 2+ in the LoVo cells and radioimmunological technique to investigate the effect of EGF on cAMP in the LoVo cells. Results\ EGF could cause a non dose-dependent increment of Ca 2+ in the LoVo cells. Forty to 60 s later, the concentration of Ca 2+ reached its peak value with the increment averaged being (143±14) %、 (147±15) %、 (146±11) %; After EGF added in 60 the medium, cAMP level in LoVo cells were rapidly decreased. Ten to 20 min later the cAMP content was decreased to its minimum, about 90 min later slowly recovered to its basic level. The mean basic values of cAMP were (48.02±4.61)×10 -8 mol/L ( n =6). Conclusion\ EGF could cause metabolism disorder of Ca 2+ and cAMP in the LoVo cells.\;
