摘要
探讨地塞米松对角膜新生血管形成及角膜内血管内皮生长因子 (VEGF)的影响。方法 :采用碱烧伤的方法制备家兔角膜新生血管模型 ,分为三组 ,每日结膜下注射地塞米松 0 .5毫克组、 1.0毫克组、对照组。分别采用宏观测量新生血管长度及生长速度 ,免疫组化染色检查 VEGF蛋白表达 ,显微镜下微血管计数方法研究角膜新生血管形成及抑制情况。结果 :实验组新生血管生长速度变慢 ,微血管数量减少 ,VEGF蛋白表达下降 ,具有统计学差异。VEGF主要表达在角膜受损区的感染细胞胞浆内 ,其出现时间与位置与角膜新生血管一致。结论 :VEGF是一种重要的角膜内新生血管形成因子 ,其变化与角膜新生血管平行 ;一定剂量地塞米松可抑制角膜新生血管 ,减少角膜内
Purposes:To elucidate the inhibitory action of Dexmethason on angiogenesis andVascular Endothelial Growth Factor(VEGF).Methods:cornea was elicited in the corneas of rabbits by Alkaline Burns,the rabbits were equally subdivided into three groups,one group was used as control,the others received subconjunctival injection of Dexmethason 0 5mg and 1 0mg for 2 weeks.To observe the occurrence and development of CNV with measurement under slit 1amp microscopy;accounting of microvessl by microscopy and immun histochemical analysis of VEGF respectively.Result:Slower CNV growth,fewer microvessl and less VEGF protein in the experiment groups than the control one.VEGF immunoreactivity was localized primarily to the inflammatory cells invading the wounded cornea.VEGF protein were temporally and spatially corralated with CNV.Conclusions:VEGF is one of important corneal angiogenic factor,some dosage of Dexmethason can decreases the express of VEGF to inhibit CNV.
