老年性白内障血、房水、晶体核中抗氧化酶和抗氧化物研究 被引量:7

A study of antioxidase and antioxidant in the blood,aqueous humor and lens nuclei of senile cataracts

摘要 目的 :了解抗氧化酶和抗氧化物之间的关系及其在老年性白内障形成中的作用。方法 :随机取符合条件的老年性白内障 44例(44眼 ) ,男 2 3例 ,女 2 1例 ,平均年龄 6 7.7岁。用黄嘌呤氧化酶法测定超氧化物歧化酶 (Superoxide dismutase,SOD)、丙二醛(Malondialdehyde MDA)用巴比妥酸反应比色法测定 ,谷胱甘肽过氧化物酶 (Glutathione- Peroxide,GSH- PX)用改进的还原型谷胱甘肽消耗法 ,Vit C用微量荧光法测定 ,Vit E用比色法测定。结果 :SOD活性在血、房水、晶体核中的活性后二者低于血中 ,但无统计学差异 (F—检验 ,P>0 .0 5 ) ;GSH— PX、 Vit C、 Vit E在血—房水和血—晶体核中的变化有显著性差异 (F—检验 ,P<0 .0 1) ;MDA在血—房水、房水—晶体核中的含量有显著性差异 (F—检验 ,P<0 .0 1)。结论 :老年性白内障的形成可能是由于房水和晶体核中抗氧化酶活性下降及抗氧化物减少和抗氧化酶之间和抗氧化物之间以及二者之间相互协调。 Objective:To investigate the relationship between the antioxidase and antioxidant,which plays a role of formation of serile cataracts.Methods:Forty four patients with senile cataracts(44 eyes)were selected at random,male for 23 cases and female for 21 cases.The activity of superoxide dismutase(SOD)was measured with xanthine oxidase method,the content of malondialdehyde(MDA)was detected with barbituric acid reaction chromometry,the glutathione peroxide(GSH PX)activity was measured with modified glutathione exhaustion assay,the content of ascorbic acid(VitC)was measured with microfluorescence method,the level of alphatocopherol(VitE)was measured with chromometry.Results:SOD activity in the aqueous humor and lens nuclei was lower than that of in the blood.It was not significantly different in them(F test,P>0 05);GSH PX activity,VitC content and VitE content were statistically of significant difference in the blood and aqueous humor as well as in the blood and lens nuclei(F test,P<0 01);MDA content was statistically of significant difference in the blood and lens nuclei as well as in the aqueous humor and lens nuclei(F test,P<0 01).Conclusion:The formation of senile catraact was due to both descending of antioxidase activity and decreasing of antioxidant content in the aqueous humor and lens nuclei(especially in the latter)and partial destruction of relationship between antioxidase and antioxidant.
