摘要
目的 检测转基因细胞分泌的表皮生长因子 (EGF)是否具有生物活性。方法 将正常表皮细胞加入培养的转基因表皮细胞于上清中培养 ,与正常表皮细胞加入培养的正常表皮细胞于上清中培养后的生长情况进行比较。通过将培养体系中加入四唑 (XTT) ,检测转基因细胞分泌的EGF是否具有刺激正常表皮细胞快速生长的生物活性。结果 实验组细胞增殖率比对照组显著增高。结论 制备的转基因表皮细胞可分泌具有刺激正常表皮细胞生长活性的EGF。
Objective To assay the bioactivity of the EGF secreted by the transfected keratinocytes. Methods The cultured normal human epidermal cells were cultured with transfected hEGF epidermal cells for 24 h; and were compared with that cultured with normal hEGF epidermal cells for 24 h, both cultured system were added XTT to test whether there was bioactivity of EGF secreted by the transfected epidermal cells. Results The epidermal cells grew more rapid than that of control group. Conclusion The transfected epidermal cells can secrete bioactive EGF.(Shanghai Med J, 2000,23∶423424)
