榄香烯乳对乳腺癌细胞株MDA-MB-231及MCF-7凋亡的影响 被引量:8

摘要 目的观察中成药榄香烯乳对乳腺癌细胞株MDA-MB-231及MCF-7凋亡的影响。方法榄香烯乳作用于MDA-MB-231及MCF-7细胞作为处理组,同时设立空白对照组;倒置显微镜观察细胞形态,结晶紫实验检测不同浓度榄香烯乳作用下的细胞生长速率,实时定量PCR检测MDA-MB-231细胞凋亡指标Caspase-3、Caspase-8、Caspase-9的表达。结果与空白对照组比较,榄香烯乳达到一定浓度时,能够有效地抑制MDA-MB-231及MCF-7细胞的生长速率(P<0.05),且呈剂量依赖性;与空白对照组比较,浓度为40μg/mL时,细胞的形态呈现典型的凋亡改变;空白对照组MDA-MB-231细胞Caspase-3、Caspase-8、Caspase-9表达量(×107拷贝量)分别为56.7±15.7、140.3±25.6、13.1±4.4,经榄香烯乳处理48 h后分别为812.2±222.6、327.8±50.1、5.7±1.5,与空白对照组比较,经榄香烯乳处理后Caspase-3、Caspase-8表达量升高(P均<0.05),Caspase-9含量无明显变化(P>0.05)。结论榄香烯乳可以有效抑制乳腺癌细胞生长的能力,并可诱导细胞凋亡。 Objective To observe the effects of elemene on apoptosis of breast cancer cell lines MDA-MB-231 and MCF-7. Methods In this study, there were two groups: the treatment group treated with elemene, and the control group. The growth rates of MDA-MB-231 and MCF-7 cells were detected by crystal violet experiment, morphological changes were observed by invert microscope, and Caspase-3, -8, -9 levels in MDA-MB-231 cells were detected by real-time PCR. Re- suits Compared with the control group, when elemene reached a certain concentration, the growth rates of MDA-MB-231 ( I〉20 ~xg/mL, P 〈 0.05) and MCF-7 ( = 160 Ixg/mL, P 〈 0.05) ceils were inhibited, with a dose-dependence; at the concentration of 40 p^g/mL, cells were changed to an apoptotic status under the microscope, and the expression levels of Caspases ( Caspase-3 : 56.7 -+ 15.7 vs. 812.2 -+ 222.6, P = 0. 025 ; Caspase-8 : 140.3 + 25.6 vs. 327.8 + 50. 1, P = 0. 032 ; Caspase-9 : 13.1 + 4.4 vs. 5.7 + 1.5, P = 0. 623) of MDA-MB-231 cells were upgraded after being treated with elemene for 48hours. Compared with the control group, the levels of Caspase-3 and Caspase-8 in the treatment group were increased ( all P 〈 0.05 ), and Caspase-9 with no obvious changes (P 〉 0.05 ). Conclusion Elemene has the capability to inhibit the growth of breast cancer cells, and the induction of apoptosis may be one of the mechanisms.
