Effects of Different Concentration Three Hormones on the Proliferation Rate of Flower Buttercup(Ranunculus asiaticus L.) Tissue Culture

摘要 [目的]研究不同浓度激素对花毛茛(Ranunculus asiaticus L.)组培增殖率的影响,为花毛茛组培苗的商业开发与应用提供参考依据。[方法]花毛茛的杂交一代(F1)种子通过无菌催芽得到无菌苗,无菌苗在基本培养基+糠基嘌呤(1 mg/L)培养基上培养驯化2代后,取增殖苗进行3因素(萘乙酸、苄氨基嘌呤、糠基嘌呤)3水平的正交试验,探索3种不同激素及其浓度对花毛茛组培增殖率的影响。[结果]花毛茛组培增殖率对萘乙酸和糠基嘌呤比较敏感,对苄氨基嘌呤反映不敏感,3激素及其浓度的最佳组合是MS+萘乙酸0.03mg/L+苄氨基嘌呤0.20 mg/L+糠基嘌呤2.50 mg/L,增殖率达到2.93,超过生产需要最低增殖率2.00水平,能够较好地满足商业生产的需要。[结论]MS+萘乙酸0.03 mg/L+苄氨基嘌呤0.20 mg/L+糠基嘌呤2.50 mg/L培养基适宜花毛茛组培增殖。 [Objective] The aim was to study the effects of different concentration hormones on the proliferation rate of flower buttercup tissue culture,and to provide a reference basis for its tissue culture seedlings' industrial development and application.[Method] Aseptic seedlings was obtained by using flower buttercup hybrid generation(F1) seed through sterile germination,aseptic seedlings cultured in the basic medium + the furfuryl purine(1 mg/L) medium was trained two generations to get proliferation seedlings which were tested three factors(NAA,benzylaminopurine,furfuryl purine) three-level orthogonal experiment,to explore the impacts of three different hormones and their concentration on the rate of proliferation of tissue culture of flower buttercup.[Result] The results showed that: the flower buttercup tissue culture proliferation rate were sensitive to NAA and furfuryl purine,not sensitive to benzylaminopurine;the best combination of the three hormones and their concentration was MS + NAA 0.03 + BA 0.20 mg/L+ KT 2.50 mg/L,the proliferation rate was up to 2.93,exceeded 2.00 level of the lowest proliferation rate the production demands,was better able to meet the commercial production needs.[Conclusion] MS+NAA 0.03 mg/L+BA 0.20 mg/L+KT 2.50 mg/L culture medium is suitable for flower buttercup tissue culture.
