慢性粒细胞白血病中X连锁凋亡抑制蛋白及其相关因子1的表达及临床意义的初步研究 被引量:1

摘要 目的初步研究慢性粒细胞白血病(CML)中X连锁凋亡抑制蛋白及其相关因子1的表达及其临床意义。方法采用实时荧光定量聚合酶链反应(RQ-PCR)检测35例慢性粒细胞白血病患者[慢性期19例,进展期16例(包括加速期5例,急变期11例)]和17例非恶性血液病组为对照组的骨髓标本中X连锁凋亡抑制蛋白(XIAP)及其相关因子1(XAF1)的mRNA表达水平,采用SPSS16.0统计学软件进行数据分析处理。结果XIAP mRNA表达水平在进展期组患者骨髓中高于慢性期组(t=4.438,P〈0.01)和非恶性血液病组(t=10.93,P〈0.01),而XAF1mRNA表达水平则低于慢性期(t=2.61,P〈0.05)和非恶性血液病组(t=10.013,P〈0.05);XIAPmRNA的表达水平在化疗后获得骨髓缓解的CML患者骨髓中显著低于化疗前(t=4.109,P〈0.01),而XAF1mRNA的表达水平则显著高于化疗前(t=5.995,P〈0.01)。结论XIAP及XAF1的联合检测及动态观察可作为CML临床辅助诊断、分期及预后判断的指标之一。 Objective To study the level of X-chromosome-linked inhibitor of apoptosis protein and its associated factor 1 and its clinical significance of Chronic myeloid leukemia (CML). Methods Used real time fluorescence quantitative polymerase chain reaction (RQ-PCR) test the level of X-chromosome-1 inked inhibitor of apoptosis protein and XIAP associated factor1 in 35 cases of CML and 17 cases of Non malignant hematologic disease,and used SPSS16.0 statistical software for data analysis. Results The expression level of XIAPmRNA in advanced group in bone marrow of patients with chronic phase (t= 4. 437,P〈0.01) was higher than that of group and non malignant blood disease group (t = 10. 93, P〈 0.01), and XAFlmRNA expression level was lower than the chronic phase (t= 2.61, P〈0.05) and non malignant hematologic disease group (t= 10. 013, P〈0. 05). The expression level of XIAPmRNA in bone marrow remission after chemotherapy for CML in bone marrow of patients with significant below before chemotherapy,and the expression levels of XAFlmRNA were significantly higher than those before chemotherapy. Conclusion Combined XIAP and XAF1 detection and dynamic observation can be used as CML clinical diagnosis,staging and prognostic indicator.
