摘要
目的:本研究以shRNA干扰CHK1在卵巢癌Skov3细胞中的表达,观察其2Gy照射后细胞凋亡情况。方法:采用shRNA技术沉默Skov3细胞中CHK1的表达,用流式细胞仪观察细胞凋亡率。结果:X射线照射后4hr和28hr,转染CHK1 shRNA的Skov3细胞的凋亡百分数为21.53%±2.51%、15.30%±2.57%,显著高于单纯转染CHK1shRNA-617组(P<0.05),显著高于转染shNC+radiation的Skov3细胞照射组(P<0.01),显著高于Control组、shNC组、radiation组(P<0.001)。结论:沉默Skov3细胞中CHK1的表达,可以明显提高卵巢癌细胞对X射线照射的凋亡敏感性。
Objectives: To observe the Skov3 cell apoptosis after 2Gy irradiation under the condition of shRNA interference with the expression of CHK1. Methods : We use shRNA vector to silence the expression of CHK1 in Skov3 cells, and observe the cell apoptosis rate with flow cytometry. Results: After 4 hr and 28 hr X - ray exposure, the cell apoptotie rates of Skov3 cells transfected with CHK1 shRNA -617 are 21.53% -+2. 51% and 15.30 -+2. 57 respectively. The rates are significantly higher than CHK1 shRNA - 617 group ( P 〈 0.05 ) , shNC + radiation group ( P 〈 0. 01 ) , control group, shNC group and radiation group( P 〈 0. 001 ). Condusion: The silent expression of CHK1 in Skov3 cells can significantly decrease the apoptosis susceptibility to X ray radiation in the ovarian cancer ceils.
