摘要
目的建立小鼠肥大细胞瘤 P815种植瘤模型 ,并对其体内诱发特异性 CTL应答机理进行初步的研究。方法经有限稀释法筛选 P815单克隆瘤系 ,用 RT- PCR及产物 DNA测序鉴定肿瘤抗原 P1A基因的表达 ;在此基础上 ,用活瘤苗免疫同系小鼠 ,经标准 5 1 Cr释放试验检测在体特异性 CTL的活性。结果筛选出了一个 P815单克隆成瘤系 P815 - F3,并鉴定该瘤存在 P1A基因的表达 ;用活瘤疫苗免疫 6只小鼠 ,在 3只体内诱发出了特异性 CTL 应答。结论本研究成功地建立了 P815肿瘤模型 ,对体内特异性抗瘤 CTL效应的诱发和检测做了初步的研究 。
ObjectiveTo establish a mouse mastocytoma P815 model and make a preliminary study on the induction of specific CTL responses in this model. MethodsWe screened a monoclone P815 cell line by limiting dilution assay (LDA),and detected whether the cell line expresses gene P1A with RT PCR and DNA sequencing. Then DBA/2 mice were immunized with the living P815 cells and monitored the specific CTL responses by 51 Cr release assay. ResultsWe have selected a cell line P815 F3 and identified the expression of P1A gene in this cell line;after immunization with the living P815 F3 cells,the anti P815AB specific CTL responses could be detected in 3/6 mice.ConclusionAfter establishing the P815 tumor model,we make a preliminary study on the induction of specific CTL responses in vivo ,proving that immunization with the living P815 cells could initiate anti P815AB specific CTL responses in about 50% mice.
