摘要
目的 :比较人正常、增生和癌变前列腺组织中PSP94和PSP5 7mRNA的表达情况。方法 :提取事故死亡的正常成年人前列腺组织及手术得到的增生和前列腺癌组织总RNA ,进行RT PCR ,其产物分别以PSP94和PSP5 7共有的外显子及PSP94外显子Ⅲ作探针进行Southernblotting分析 ;RT PCR产物经克隆后进行序列测定。结果 :三种前列腺组织中均有PSP94和PSP5 7mRNA的表达 ;人正常前列腺组织中的PSP94及PSP5 7的基因序列与增生前列腺组织和癌变前列腺组织中的完全一致。结论 :PSP5
Objective:Analysize and compare the expression of human PSP94 and PSP57 mRNA in prostatic tissues from normal adults, patients with prostate carcinoma and benign prostatic hyperplasia (BPH).Methods:Extract total RNA in prostatic tissues taken by operation from normal adults died of accident, patients with prostate carcionma and BPH, then do reverse transcriptase polymerase chain reaction (RT PCR). RT PCR products are analysized by sequence determination and Southern blotting using specific exon Ⅱ of PSP94 and PSP57 and exon Ⅲ of PSP94 as probes.Results:The PSP94 and PSP57 mRNA are both found in the above three kinds of prostatic tissues. The cDNA sequences of PSP94 and PSP57 in normal prostatic tissue are consistent with that in carcinoma and hyperplasia tissues. Conclusion:PSP57 is a non specific product of PSP94 mRNA in prostatic tissues from patients with prostate carcinoma and benign prostatic hyperplasia.
