植物源抗病毒剂VFB提取工艺的优化

Studies on Extraction Process Optimization of Botanical Virucide VFB

为优化植物源抗病毒剂VFB加工工艺,采用单因子试验和正交试验研究VFB加工用植物材料的提取时间、提取次数和料液比等工艺参数,并以心叶烟和曼陀罗为寄主材料,采用半叶枯斑法测定VFB抗TMV的活性。结果表明,VFB最佳提取工艺参数为提取时间20h,提取5次,m(植物材料)∶V(水)=9∶1;生测表明,优化后的VFB在50倍液剂量下,钝化TMV病毒粒子5 min,其抑制率分别达到83.33%和88.00%,较旧配方分别提高39.33%和48.80%;处理6h时,对TMV初侵染的抑制率分别达到74.20%和77.15%,较旧配方分别提高20.20%和26.50%;处理6h后,对TMV复制增殖的抑制率分别达到66.67%和69.00%,较旧配方分别提高29.34%和29.60%。可见,经提取工艺优化后,VFB的抗TMV活性有明显地提高。

The objective of this research was to optimize the technological parameters for botanical vi rucide VFB by the single factor experiment and the orthogonal trail. These parameters included the extraction time of plant material, extraction times, ratio of solvent and plant material. In addition, its activity against TMV was investigated by local lesion and the lesion hosts were Nicotiana glutinosa I.. and Datura stramonium Linn.. The results indicated that the most appropriate parameters was as following,the ratio of solvent and plant material is 9 ： 1,the extraction times is 5, each time for 4 h. The optimized VFB showed significant activity against TMV under the circumstance of 50 times the dosage and mixing VFB with TMV for 5 rain,the inhibition rate were 83.33% and 88.00% ,increased by 39.33% and 48.80% respectively. The inhibiting rate of infectivity to TMV 6 h after application were 74.20% and 77.15%,increased by 20.20% and 26.50% respectively. The inhibition rates were 66.67% and 69.00% in the experiment of inhibiting multiplication on TMV after 6h application,in- creased by 29.34% and 29.60% respectively. In summary,the anti-TMV activity of optimized VFB had been improved significantly.
