香芪汤及其有效成分对LPS诱导的巨噬细胞MAPK信号通路的影响 被引量:6

Effects of Xiang-Qi-Tang and its active components on MAPK pathway of macrophage induced by LPS

摘要 为揭示中药香芪汤及其有效成分的体外抗炎作用机理,利用ELISA法研究了不同质量浓度(0.2、1、5mg/L)的香芪汤对细胞因子TNF-α分泌的影响,并运用Western blot法检测了香芪汤及其有效成分黄芪甲苷、α-香附酮和穿心莲内酯对ERK1/2和JNK1/2的影响。结果显示,1、5mg/L质量浓度的香芪汤能够极显著抑制RAW264.7巨噬细胞TNF-α的分泌(P<0.01),香芪汤及其有效成分能够极显著降低JNK1/2和ERK1/2的磷酸化水平(P<0.01)。由此推论,中药香芪汤及其有效成分可能通过抑制炎症因子的分泌,降低MAPK信号通路的活性发挥其抗炎作用。 To reveal the anti-inflammation mechanism of Xiang-Qi-Tang and its active components of Chinese herbal formula in vitro, We determined different concentrations of Xiang-Qi-Tang(0. 2,1,5 mg/L) on the influence of secretion of TNF-α by ELISA as well as the influence of Xiang-Qi-Tang and its active components including astragalosideIV, α-cyperone and andrographolide on the ERK1/2 and JNK1/2 via Western blotting. The results showed that 1 and 5 mg/L of Xiang- Qi-Tang can significantly inhibit the secretion of TNF-α of RAW264.7 macrophages(P〈0.01), meanwhile,Xiang-Qi-Tang and its active components can memorably reduce the phosphorylation level of ERK1/2 and JNK1/2(P〈O. 01). The results indicated that Xiang-Qi-Tang and its active components may play an anti-inflammation activity by mean of the reduction of mediator of inflammation and the attenuation of the activation of MAPK signal pathway.
