利用DGE对黄肉猕猴桃果实着色过程中相关差异基因表达的解析 被引量:7

摘要 【目的】构建数字基因表达谱(DGE),寻找调控黄肉猕猴桃果实呈色的潜在关键基因。【方法】以中华猕猴桃(Actinidia chinensis Planch)黄肉品种‘金丰’为试材,利用Illimina/Hisseq测序技术构建其果实发育中3个不同转色期DGE文库,以转录组数据为参考数据库,进行GO功能富集以及Pathway功能富集分析。【结果】结果表明,DGE文库构建转色前期、转色期和成熟期分别获得4 724 009,4 565 159和4 626 730个标签。转色期与转色前期之间差异表达基因的个数为2 641个,成熟期与转色前期有差异表达的基因个数是2 671,成熟期与转色期之间有1457个基因有表达差异。通过GO功能富集及Pathway功能富集分析,分别获得了102条叶绿素Unigenes与111条类胡萝卜素Unigenes,其中12条叶绿素Unigenes及14条类胡萝卜素Unigenes在果实的3个不同转色期有显著差异表达(P<0.05)。【结论】黄肉猕猴桃呈色的关键基因可能存在于具有显著差异表达的12条调控叶绿素合成以及14条调控类胡萝卜素合成的Unigenes之中。 [Objective]The objective of this study is to construct the Digital Gene Expression(DGE) tag profiling and find out the potential genes related to controlling the fruit color in yellow flesh kiwifruit. [ Method ]Taking yellow flesh cultivar from A ctinidia chinensis Planch., named ' Jinfeng' as tested mate- rials, and three DGEs of different color-turning stages by Illimina/Hisseq sequencing technique were con- structed and the enrichment analysis of Gene Ontology (GO) and Pathway function was conducted based on the data of transcriptome.[Result] The results showed that there were 4 724 009,4 565 159 and 4 626 730 tags respectively, which obtained from DGE during the color-turning pre-phase, color-turn- ing phase and maturing phase. And 2 641 genes performed discrepant expression in color-turning pre- phase and color-turning phase ; 2 671 genes performed discrepant expression in color-turning pre-phase and mature phase ; 1 457 genes performed discrepant expression in color-turning phase and mature phase. In addition, the enrichment analysis of GO and Pathway function indicated that significant discrepancy existed in the 12 of the 102 Unigenes on chlorophyll synthesis and 14 of the 111 Unigenes on caroteniod synthesis in the three color-turning phases of 'Jinfeng' kiwifruit fruit. [ConclusionlThis study suggested the key genes controlling fruit color in kiwifruit with yellow flesh might present in the 12 Unigenes related chlorophyll synthesis and 14 Unigenes related caroteniod synthesis which held significant discrepancy in gene expression.
