基于SSR标记的早花柠檬亲缘关系分析 被引量:12

摘要 【目的】目前对早花柠檬的遗传背景、起源分类研究尚属空白,为了探究早花柠檬与其他柑橘种质间的亲缘关系,【方法】利用筛选的11对SSR多态性引物对早花柠檬及16份柑橘属及其近缘属材料进行DNA扩增分析。【结果】结果共检测到92个等位变异,平均每对引物检测到8.5个。17份柑橘属及其近缘属材料的遗传相似系数变化在0.59~1.00。以相似系数0.65为阈值,UPGMA聚类分析可以将17份材料分为4个类群。早花柠檬、枸橼、尤力克柠檬、北京柠檬、费米耐劳柠檬、粗柠檬等一起构成了第2类群,其中早花柠檬与枸橼的遗传相似系数最高,为0.87。【结论】该结果为研究早花柠檬的起源、进化及分类提供了重要线索。 [Objective]The objective of the research is to study the phylogenetic relationship between early flowering lemon and other accessions of citrus and its related genera. [Method] In this study, a total of 11 pairs of polymorphic Simple Sequence Repeat (SSR) markers were detected in early flowering lemon and 16 accessions of citrus and its related genera. [Result]Based on SSR data, cluster analysis was performed via un-weighted pair group method analysis (UPGMA) method using NTSYS-PC 2.10. These primers gen- erated 93 alleles (2-15 alleles per locus) with a mean value of 8.5 alleles per locus. Cluster analysis showed that the 17 materials can be divided into four main groups at the coefficient of 0.65. In the second group, the early flowering lemon and citron were separated from the subgroup including Eureka lemon, Femminello lemon, Mayor lemon and Rough lemon at the similarity level of 0.71. Early flowering lemon and citron were clustered into a subgroup with the coefficient of 0.87. [Conclusion]The present conclusion can be used for citrus systematic taxonomy and genetic breeding.
