摘要
目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合放射线对Lewis肺癌细胞增殖及凋亡的影响。方法采用MTT法检测不同浓度TRAIL对Lewis肺癌细胞增殖的影响;流式细胞仪检测TRAIL、X射线及两者联合对Lewis细胞凋亡率及细胞周期的影响。结果 TRAIL对Lewis细胞的体外抑制作用与浓度相关(F=9.017~12.465,P<0.05)。TRAIL与X射线联用能够使Lewis细胞的凋亡率及G2/M期比例提高,与对照组及单用TRAIL组和单用X射线组相比,差异有统计学意义(F=203.175、149.276,P<0.05)。结论 TRAIL可抑制Lewis细胞的增殖,并与浓度相关。TRAIL与放射线联用能够提高肿瘤细胞的凋亡率及细胞周期中G2/M期的比例。
Objective To investigate the effect of combined tumor necrosis factor-related apoptosis inducing ligand(TRAIL) with radiation on cell multiplication and apoptosis of Lewis lung cancer cells.Methods By utilizing MTT method and flow cytometry to respectively detect the effects of different-concentration TRAIL on multiplication of Lewis lung cancer cells,and influence of combined TRAIL with X-ray on the cell apoptosis and the cell cycle.Results The inhibition of TRAIL on Lewis cells-in vitro-was related to the drug concentration(F=9.017-12.465,P<0.05).The combined use of TRAIL and X-ray irradiation could raise Lewis cell apoptosis and G2/M period of the cell cycle,compared with the control group and TRAIL or X-ray irradiation alone,the differences were significant(F=203.175,149.276;P<0.05).Conclusion TRAIL can inhibit the proliferation of lung cancer cells,which is TRAIL-concentration-related.Combining TRAIL with radiation can raise the apoptosis of cancer cells and ratio of G2/M in the cell cycle.
