摘要 目的建立胃癌癌前病变细胞模型。方法使用与胃癌相关的亚硝胺类化合物N-甲基-N′-硝基N-亚硝基胍(MNNG)对永生化的人胃黏膜上皮细胞系GES-1进行恶性转化实验,得到转化后的MC细胞。比较观察GES-1细胞和MC细胞的形态,同时检测两种细胞中神经钙黏附素(N-cadherin)与磷酸化信号传导和转录激活因子3(P-STAT3)的表达。结果与GES-1细胞比较,MC细胞形态以及生长特性均发生了改变。随传代的延续,细胞呈现典型的"岛样"克隆生长。胃癌组织相关蛋白N-cadherin、P-STAT3的表达量增加。结论本研究成功构建了胃癌癌前病变细胞模型。 Objective To establish a cell model of gastric precancerous lesions.Methods By using gastric-cancer-related N-methyl-N′ nitro-N-nitrosoguanidine nitroguanidine(MNNG),malignant transformation experiments were done in human gastric epithelial cell line GES-1,and converted MC cells were then obtained.The morphology of GES-1 cells and MC cells were observed and compared,the expressions of N-cadherin and P-STAT3 in the above two cells were detected.Results Compared with GES-1 cells,the morphology and growth characteristics of MC cells were changed.With the continuous passage,the cells showed a typical 'island-like' cloning growth.The expressions of gastric-cancer-related N-cadherin and P-STAT3 increased.Conclusion A cell model of gastric precancerous lesions has been successfully constructed by this study.
