摘要
目的探讨高压氧预处理对大鼠肝脏缺血再灌注损伤的影响。方法 40只雄性SD大鼠随机分为4组,假手术组(SO组);缺血再灌注组(IR组);高压氧预处理1组(HBO1组);高压氧预处理2组(HBO2组),每组10只。测定4组大鼠经处理后血清丙氨酸氨基转移酶(ALT)和天冬酸氨基转移酶(AST)以及各组大鼠经处理后肝脏组织学变化。结果 HBO2组及IR组ALT、AST水平明显高于SO组(P<0.05);HBO2组ALT、AST水平高于IR组(P<0.05);与HBO1组比较,HBO2组及IR组ALT、AST浓度差异均有统计学意义(P<0.05);HBO1组ALT、AST水平虽有上升,但与SO组比较差异无统计学意义(P>0.05)。HBO1组肝脏组织、小叶及汇管区结构清晰,可见肝细胞轻度水肿及瘀血;IR组肝小叶及汇管区可见轻度炎细胞浸润,片状肝细胞水肿,少数明显内空泡样变,肝组织瘀血,肝细胞坏死轻微;HBO2组肝脏小叶结构存在,肝小叶及汇管区可见大量炎细胞浸润,肝细胞坏死严重,HBO2组肝细胞水肿、坏死范围大于IR组。结论适量的高压氧预处理对肝脏缺血再灌注损伤有保护作用,而过量高压氧预处理,5d加重大鼠肝脏缺血再灌注损伤。
Objective To investigate the effect of hyperbaric oxygen preconditioning on ischemia-reperfusion injury in liver of rats.Methods 40 male SD rats were randomly divided into 4 groups,sham operation group(SO group),ischemia reperfusion group(IR group),hyperbaric oxygen preconditioning 1 group(HBO1 group),hyperbaric oxygen preconditioning 2 group(HBO2 group),with 10 rats in each group.The activities of alanine aminotransferase(ALT) and aspartic aminotransferase(AST) in serum were detected,and the histological changes of liver after treatment were observed for all the rats in the four groups.Results The serum levels of ALT and AST in HBO2 group and IR group were significantly higher than those in SO group and IR group(P〈0.05),and the serum levels of ALT and AST in HBO2 group were significantly higher than those in IR group(P〈0.05).There were significant differences in serum levels of ALT and AST between HBO1 group and HBO2 group or IR group(P〈0.05).The levels of ALT and AST in HBO1 group were increased,but there were no significant differences between HBO1 group and SO group(P〈0.05).The liver tissue,lobular and portal structure were clear in HBO1 group,with liver cell mild edema and ecchymosis being visible;in group IR the lobular and portal showed mild inflammatory cell infiltration,flaky edema of liver cells,a few significant intra vacuolar degenerations of liver tissue,liver tissue haemostasis and mild liver cell necrosis;in HBO2 group the liver lobule structure existed,a lot of inflammatory cell infiltrations were visible in hepatic lobule and periportal region,with severe liver cell necrosis,and the extent of necrosis and edema of liver cells in group HBO2 was greater than that in group IR.Conclusion The suitable hyperbaric oxygen preconditioning can relieve ischemia-reperfusion injury of rat's liver,howeve,overdosage hyperbaric oxygen preconditioning may aggravate ischemia-reperfusion injury of liver of rats.
