姜黄素对局灶脑缺血再灌注损伤的神经保护作用及对新生神经细胞增殖的影响 被引量:5

摘要 目的:观察姜黄素对大鼠局灶性脑缺血再灌注损伤后的神经保护作用,以及姜黄素对新生神经细胞增殖的影响。方法:建立SD大鼠大脑中动脉缺血再灌注损伤的模型,分空白对照组、假手术组、模型对照组、姜黄素给药组;在脑缺血再灌注损伤后1 h腹腔注射姜黄素,连用7 d后处死,观察神经功能缺损评分、梗死体积的变化,免疫组化检测梗死侧侧脑室下区5-溴-2-脱氧尿嘧啶核苷(BrdU)标记的新生神经细胞。结果:7 d后姜黄素给药组神经功能缺损评分呈现下降的趋势;脑梗死体积较模型对照组显著减少(P<0.01);BrdU标记的新生神经细胞数较模型对照组显著增多(P<0.01)。结论:姜黄素对局灶性脑缺血再灌注损伤具有神经保护作用,能促进BrdU标记的新生神经细胞增殖。 Objective: To observe the neuroprotection and neurogenesis effect of curcumin on focal ischemia reperfusion injury. Methods: The focal ischemia reperfusion injury model was establisbed by middle cerebral artery occlusion and rats were divided into four groups: normal control group, sham-operated group, model control group and curcumin group. Curcumin group was treated with curcumin (300 mg/kg) after ischemic perfusion 1 h last for 7 d by intraperitoneal injection and the other groups were injected with equal coil oil 7 d too. Neurobehavioral deficit, infarction volume, BrdU's expression were studied in our experiment with TTC staining and immunofluo- rescence technique. Results: As compared with model group, curcumin group's neurobehavioral deficit has a descend trend; infarction volume was decreased markedly and there was a statistical difference between model group and curcumin group (P〈0.01); BrdU-positive cells were conspicuously higher than model control group (P 〈0.01). Conclusion: Curcumin has neuroprotection and neurogenesis effect on focal ischemia repel-fusion injury.
