人参可溶性糖提取过程中两种除蛋白质方法的比较 被引量:1

摘要 采用Sevage法和乙醇沉淀法分别处理人参可溶性糖粗提液,以人参可溶性糖保留率和蛋白质去除率为参数通过正交试验和单因素试验优化两种方法的除蛋白反应条件。结果表明,Sevage法去除人参可溶性糖粗提液中蛋白质的最优条件为:溶液配比1:4,反应时间40 min,Sevage溶液与样品液比例为1:3,处理样品液1次;乙醇沉淀法去除人参可溶性糖粗提液中蛋白质的最优条件为:乙醇浓度30%,加热温度70℃,加热时间1.5 h。在去除人参可溶性糖粗提液中蛋白质的试验过程中,Sevage法优于乙醇沉淀法,适用于实验室中纯化人参中的糖类,但乙醇沉淀法在大量处理人参可溶性糖溶液时更便于操作。 The Sevage method and ethanol precipitation treated separately crude extracts of ginseng soluble sugar, ginseng soluble sugar retention and protein removal rate parameters were optimized by orthogonal experiment and single factor experiment two methods in protein-removing reaction conditions The results showed that the optimal conditions of Sevage method for removing protein as follows: the solution ratio was 1:4, the reaction time was 40 min, Sevage solution with the sample liquid ratio was 1:3 and deal the sample solution once. The optimal conditions of ethanol precipitation for removing protein as follows: ethanol concentration was 30%, the heating temperature was 70~C, the heating time was 1.5 h. Sevage method wass more user-friendly than ethanol precipitation for laboratory purified ginseng sugars, but ethanol precipitation was easy to operate in a large number of processing ginseng soluble sugar solution.
