摘要
采用Sevage法和乙醇沉淀法分别处理人参可溶性糖粗提液,以人参可溶性糖保留率和蛋白质去除率为参数通过正交试验和单因素试验优化两种方法的除蛋白反应条件。结果表明,Sevage法去除人参可溶性糖粗提液中蛋白质的最优条件为:溶液配比1:4,反应时间40 min,Sevage溶液与样品液比例为1:3,处理样品液1次;乙醇沉淀法去除人参可溶性糖粗提液中蛋白质的最优条件为:乙醇浓度30%,加热温度70℃,加热时间1.5 h。在去除人参可溶性糖粗提液中蛋白质的试验过程中,Sevage法优于乙醇沉淀法,适用于实验室中纯化人参中的糖类,但乙醇沉淀法在大量处理人参可溶性糖溶液时更便于操作。
