摘要
在前期形态学分类及大量RAPD分析的基础上,选取在RAPD扩增45号哈茨木霉菌株获得的1条特异性片段,将其成功转化为1个稳定的SCAR标记。结果表明,该SCAR分子标记具有种的特异性,可将哈茨木霉属与其他木霉属分开。利用该标记对供试的45个木霉菌菌株进行PCR特异性扩增时,哈茨木霉菌菌株均能扩增出1条779 bp的DNA条带,不属于哈茨木霉的菌株则无扩增产物。
Based on the previous morphological classifications and results of RAPD for 45 Trichoderma strains, the special amplification products from RAPD in Trichoderma strain No.45 were sequenced. A pair of primers were designed and successfully converted into SCAR marker. The results showed that the SCAR marker could distinguish Trichoderma harzianum from other Trichoderma strains. Using the special primers for specific amplification of 45 Trichoderma strains tested, T. harzianum strains could be amplified by an 779 bp special band, and the other strains could not find the amplified products.
