Effects of Chen Weiliang’s Traumatic Oil on Cytokine IL-1β and PGE_2 in Rats Acute Injured Tissue

摘要 目的探讨陈渭良伤科油对急性软组织损伤中炎性细胞因子白介素-1β(IL-1β)及前列腺素2(PGE2)含量的影响,进一步评价其作用机理。方法 48只成年SD大鼠随机分为正常对照组、模型组、万花油组及陈渭良伤科油组(简称伤科油组)4组,每组12只。模型组、万花油组、伤科油组均选取大腿外侧臀肌处使用自制打击器打击复制急性软组织损伤模型后,分别外用石蜡油、跌打万花油、陈渭良伤科油,共3 d。同时分批处死大鼠取病变组织进行匀浆后进行IL-1β、PGE2含量的测定。结果伤科油能使损伤局部IL-1β的含量降低,并在伤后12 h即开始发挥作用,而万花油治疗作用出现在受伤后48 h;伤科油组能使损伤组织局部PGE2的含量降低(P<0.05),并在伤后12 h发挥作用,72 h时PGE2的含量接近正常对照组水平。万花油组PGE2含量各时间段无明显变化。结论伤科油可通过减少损伤组织炎症因子的含量起到抗炎镇痛作用。 Objective To investigate Chen Weiliang's Traumatic Oil(CTO) on the contents of cytokines interleukin-1 beta(IL-1β) and prostaglandin E2(PGE2) in rats acute injured tissue, and to explore the mechanism. Methods Forty-eight male SD rats were divided into four groups, blank control group, model group, CTO group and Wanhua Oil group, 12 rats in each group. Except for the blank control group, rats in the other groups were stroked on lateral gluteus by self-made apparatus to induce acute tissue injury. After the modeling, model group, CTO group and Wanhua Oil group were given external application of petroleum, CTO and Wanhua Oil for 3 days, respectively. SD rats in the four groups were executed in batches, and the contents of IL-1β and PGE2 in the soft tissue homogenate were detected. Results CTO had an effect on decreasing the IL-1β content in the focal injured tissue 12 hours after injury, and Wanhua Oil had the effect 48 hours after injury. PGE2 content was decreased in the injured tissue of CTO group 12 hours after injury, and the effect lasted over 72 hours. Wanhua Oil had no such effect. Conclusion It is demonstrated that decreasing the release of inflammatory factors in injured tissue is one of the anti-inflammatory and analgesic mechanisms of Chenweiliangg' s Traumatic Oil.
