摘要
为对大麦种子的真伪进行有效监控,防止假冒伪劣品种进入市场,也为新品种登记测试以及种质资源遗传多样性分析提供参考,采用SSR标记技术对29个大麦DUS测试标准品种进行了遗传多样性分析和指纹图谱的构建。利用筛选出的28对有多态性的SSR引物共扩增出125个等位基因,每对引物可扩增出3~7个等位基因,平均4.46个等位基因,每对引物在29个品种中的PIC值为0.25~0.81,平均为0.62。聚类分析表明,29个大麦DUS测试标准品种可聚成4类,具有较远的亲缘关系,这些材料首先按照穗棱型进行聚类,表明不同穗棱型品种间存在较大遗传差异。构建了29个大麦DUS测试标准品种在28个SSR位点上的指纹图谱。
To provide reference for monitoring the authenticity of barley varieties,preventing counterfeit and inferior varieties in the market,registration test of new varieties and genetic diversity analysis of germplasm,this study analyzed the genetic diversity among 29 barley standard varieties in DUS testing and built their DNA fingerprint map by 28 pairs of SSR markers with good polymorphism.28 pairs of SSR primers with polymorphism were detected and 125 alleles were measured in total.Each pair of primers could detect 3~7 alleles with 4.46 alleles in average.The polymorphism information content(PIC) among the primers got the highest of 0.81 and the lowest of 0.25,with the average of 0.62 among the 28 pairs.Clustering analysis showed that these varieties could be clustered into 4 groups.The results showed the SSR genetic differences among 29 materials were abundant,groups were structured in priority according to row-type,which showed that there were large genetic differences between varieties with different row-types.
