摘要
目的分析吉林省延边朝鲜族自治州(延边州)流行的肠道病毒71型(EnterovirusType71,EV71)分离株的基因特征。方法对2009~2010年延边州手足口病(Hand,Footand Mouth Disease;HFMD)来源的5株EV71分离株进行逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)扩增VP1编码区,测序并使用Bioedit和Maga4生物信息学软件进行基因特征分析。结果延边州5株EV71分离株均为C4a基因亚型,其VP1编码区核苷酸序列具有较高的同源性(99.3%~100%),在基因树上形成紧密的一簇。延边州的5株分离株与安徽省阜阳市2008年流行株具有最近的基因亲缘关系,核苷酸同源性高达99.3%~99.4%。结论延边州2009~2010年EV71分离株均属于C4a亚型,至少有一个EV71传播链在延边州持续传播,引起延边州HFMD的流行。需要加强EV71分子流行病学监测,掌握本地流行的EV71基因型数据,并及时发现输入基因型。
Objective To analyze the genetic characteristics of enterovirus type 71 (EV71) strains isolated from suspected patients of hand, foot and mouth disease (HFMD) in Yanbian korean autonomous prefecture(Yanbian) of Jilin province. Methods The VP1 genes coding region of 5 EV71 strains from HFMD in Yanbian in 2009-2010 were amplified by reverse transcription-polymerase chain reaction (RT-PCR) and were sequenced. Gene characteristics were analyzed by Bioedit and Mega 4 software. Results All the 5 strains belonged to C4a subgenotype. The homology of nucleotide sequences in VP1 coding region of the 5 strains was 99.3 % -100%, and they formed a closed cluster. The 5 strains have the nearest phylogenetic relationship with that isolated in Fuyang prefecture of, Anhui province in 2008, with a homology of nucleotide of 99.3 % -99.4%. Conclusions 5 strains of EV71 isolated in Yabian from 2009 to 2010 were C4a subgenotype. At least one EV71 transmission chain was circulating and caused the HFMD epidemic in Yanbian. We should strengthen the EV71 molecular epidemiological surveillance, grasp the genotype data of local circulating EV71 and promptly find the imported genotype.
