摘要 选择Plackett-Burman设计和响应面优化相结合的方法,对米曲霉CGMCC 2.0951和热带假丝酵母CGMCC 2.118混合固态发酵玉米皮制备菌体蛋白质饲料的培养基组成进行了优化。二水平PB设计对影响玉米皮发酵的8个因素进行筛选,得出影响最大的3个因素:麸皮、尿素和K2HPO4;再通过响应面分析法对这3个因素进行优化,得出最适添加量分别为麸皮7.63%、尿素1.54%和K2HPO41.16%。优化后的玉米皮培养基经过混菌发酵,其真蛋白质含量可达到12.54%,较发酵原料提高了1.65倍。 Two stage statistical design was used to optimize microbial biomass protein production from Aspergillus oryzae CGMCC2.0951 and Candida tropicalis CGMCC2.118 under solid-state fermentation.Initially,Plackett - Burman designing (PB) was used for the selection of crucial production parameters.Wheat bran,urea and K2HPO4 were found to be the crucial factors for the microbial biomass protein production.Crucial variables were further processed through Box- Behnken of response surface methodology (RSM) to maximize the true protein yield.Three factors were investigated at three different levels to study their influence on true protein production.Statistical approach showed that the optimum conditions for true protein production (wheat bran 7.63%,urea 1.54%and K2HPO4 1.16%) resulted in 1.65-fold increase in true protein yield (12.54%) over raw materials.
