植物油中角鲨烯的提取与高效液相色谱法分析 被引量:20

摘要 建立了硅胶柱提取,高效液相色谱法分析植物油中角鲨烯的方法。样品用石油醚溶解,过硅胶柱净化,经C18柱分离,紫外检测(210 nm)。结果表明,角鲨烯与其他杂质峰得到良好分离,在2~1 000 mg/L浓度范围内,峰面积与浓度呈良好线性,线性相关系数(R2)0.998。在大豆油中添加5.0、100、500和1 000mg/kg水平的角鲨烯,平均加标回收率为82.4%~89.3%,相对标准偏差小于7.1%(n=8),方法测定低限(LOQ)5 mg/kg。本方法准确、灵敏、可靠,已应用于实际样品分析。 A method for the extraction of squalene in vegetable oils by silica gel column and determination by high performance liquid chromatography( HPLC )was established. Vegetable oil samples were dissolved with petroleum ether,cleaned up with a silica gel column ,separated with C18 column and determined by ultraviolet detection at 210 nm. The results showed that using this method, squalene and other impurity peaks obtained good separation. In 2 - 1 000 mg/L concentration range, the peak area against concentration showed good linear relationship. The linear cor- relation coefficient(2)was 0.998. Soybean oil was added with squalene at 5.0,100,500 and 1 000 mg/kg, respectively. The average recoveries than 7.1% ( n = 8 ). The limit ranged from 82.4% to 89.3% , and the relative standard deviations(RSDs) were less of quantification could achieve 5 mg/kg. This method was accurate, sensitive and reliable, which has been applied to the real sample analysis.
