摘要
为了研究奶牛乳腺葡萄糖转运蛋白1(GLUT1)对葡萄糖的转运作用,试验采用荧光定量PCR技术、Western-blot和激光扫描共聚焦显微技术检测奶牛乳腺发育不同时期的表达情况。结果表明:GLUT1 mRNA和蛋白在不同发育时期的奶牛乳腺中均有表达,并且表达趋势相似,泌乳期持续高表达,泌乳140天达到峰值;GLUT1主要定位于乳腺上皮细胞基底侧细胞膜和顶膜。说明GLUT1可以将细胞外葡萄糖转运至细胞内。
To investigate the transfer effect of glucose transporter - 1 protein ( GLUTI ) on glucose in dairy cow mammary gland, the real - time PCR technique, Western blotting and laser scanning eonfoeal microscopy, were used to determine the expressions in dairy cow mammary gland at different developmental periods. The results showed that GLUT1 mRNA and protein were expressed in the dairy cow mammary gland at different development periods, and their expression showed similar trends, which sustained high expression during lactation and peaked at 140 d of lactation. GLUTI was mainly located in basolateral and apical membranes of mammary epithelial cells. It indicates that GLUTI can transport extracellular glucose into the cells.
