生长抑素DNA疫苗表达产物在小鼠组织细胞中的分布 被引量:1

摘要 为了揭示无抗性筛选生长抑素DNA疫苗表达产物的分布,试验用非抗性筛选生长抑素DNA疫苗(pGS/2SS-M4GFP-asd)肌肉注射7周龄的雄性昆明小鼠,24 h、48 h、5 d、7 d、9 d、11 d后分别采集脾脏、肝脏、肌肉等组织器官,经免疫组织化学染色,检测DNA疫苗表达产物在小鼠各组织器官中的分布情况。结果表明:DNA疫苗表达产物在免疫后不同时间出现在宿主肌肉组织的中性粒细胞、巨噬细胞、成纤维细胞、脾脏巨噬细胞及肝细胞中,在心脏、肺脏、肾脏各组织的细胞中尚未检测到生长抑素(SS)阳性信号。说明该DNA疫苗能稳定地在宿主体内表达,为开发高效的促进动物生长的DNA疫苗提供重要的理论基础。 To reveal the distribution of the expression products of non-resistance screening somatostatin (SS) DNA vaccine in tissues and cells, the SS DNA vaccine( pGS/2SS- M4GFP -asd)was injected into the male Kunming mice of 7 weeks old in the study. The tissues of heart, liver, spleen, lung, kidney, muscle were taken after 24 h, 48 h, 5 d, 7 d, 9 d and 11 d, respectively, and then embedded in paraffin, stained with immunohistochemistry technique to detect the distribution of the expression products of the DNA vaccine in the tissues and organs of the mice. The results showed that the expression products of the SS DNA vaccine appeared in neutrophils, macrophages, flbroblasts of muscular tissue, spleen macrophages or hepatocytes at different periods after immunization in the mice. However, the SS positive signal was not yet detected in the tissues of the heart, lung and kidney. The results demonstrate that the DNA vaccine can be expressed stably in mice, providing an important theoretical basis for the development of efficient DNA vaccines to promote animal growth.
