摘要
本文应用单克隆抗体固相致敏红细胞吸附技术(SPASE)检测血液中的HBsAg。对该方法的实验条件和影响因素进行了研究,并改进了抗体的固相方法,使检测HBsAg时可以不稀释血清而直接检测。通过219份临床标本的检测,证明该方法特异性和敏感性均高于反向间接血凝法(RPHA),且操作简便,快速,节省人力物力,更适合于基层实验室推广应用,
A new method, Solid-phase Adsorption of Sensitized Etythrocytes (SPASE), for detecting HBsAg from sera was established and the conditions influencing the performances of the method were investigated. The way for solid-phase adsorption of antibodies onto the surface of plexiglass microtiter plate was improved. It was showed that the sera to be examined could be tested in original soluion without nonspecific reactions. The results of examination of 219 specimens from clinic by SPASE and RPHA ( reverse passive hemagglutination ) simultaneously demonstrated that the sensitivity and specificity of the former were significantly higher than those of the latter.We consider that SPASE is especially suitable for basic-level clinical laboratories.
