红树林来源真菌PJX-41次级代谢产物及其抗肿瘤活性研究 被引量:3

摘要 对1株红树林来源的海洋真菌PJX-41发酵产物中的抗肿瘤成分进行分离和鉴定。采用溶剂萃取、硅胶柱色谱及制备HPLC等分离手段得到6个喹唑啉酮生物碱类化合物,利用波谱学手段(核磁共振、有机质谱、X-Ray单晶衍射等)鉴定了单体化合物分别为quinadoline B、glyantrypine、lapatin A、tryptoquivaline、deoxytryptoquivaline和deoxynortryptoquivaline。以磺酰罗丹明B(SRB)法和四甲基偶氮唑盐(MTT)法评价了化合物的抗肿瘤活性,结果表明:lapatin A对A-549和Hela细胞具有中等抗肿瘤活性,tryptoquivaline对Hela细胞具有中等抗肿瘤活性,其他化合物没有活性。这6个化合物为首次从该菌中分离得到。 The anti-tumor activity components were isolated and identified from the fermentation product of the fungus Cladosporium sphaerospermum derived from marine mangroves. Six quinadoline alkaloids compounds were obtained by solvent extraction, silica gel column chromatography and preparative HPLC. Their structures were identified as quinadoline B, glyantrypine, lapatin A, tryptoquivaline, deoxytryptoquivaline and deoxynortryptoquivaline respectively by spectroscopy methods such as NMR, mass spectrometry and X-Ray. The cytotoxie activity of the compounds was evaluated by SRB and MTT methods, and the results showed that lapatin A has weakly anti-tumor activity aginst both A-549 and hela ceils, tryptoquivaline has weakly activity against hela cell, and other compounds have no activity. This is the first time to get compounds from this fungus.
