摘要
利用IPTG作为诱导剂进行鸡β-防御素-3基因重组蛋白的表达,探讨工程化生产的可能性。分别对影响重组蛋白表达的因素(IPTG浓度、诱导时机、诱导时间、诱导温度和溶氧量)进行优化,最终得到最佳的IPTG诱导条件:IPTG的终浓度为0.6mmol/L,诱导起始吸光度A600为0.6,最适溶氧量为150mL三角瓶装50mL培养基,且在33℃下诱导4h重组蛋白表达量达到最大。该方法为工程化生产提供良好的试验依据。
To study the expression of the recombinant protein of chicken Defensin-β-3 induced with IPTG,and explore the possibility of engineering production,the effects of the concentration,induction time,induction temperature and the dissolved oxygen of IPTG on the expression of the recombinant product were compared to determine more optimal conditions for IPTG induction.For the production of recombinant Gal-3,the expression level of induction product was better with 0.6 mmol/L IPTG at A600 0.6,the appropriate load is 50 mL culture medium in 150 mL flask and 33 ℃ induce 4 h.This study provides a good experimental basis for the large production of Gal-3.
