杂交瘤细胞系产生单克隆抗体稳定性研究

摘要 大文对10株乙肝表面抗原(HBsAg)的McAbs细胞系在BALB/c小鼠体内连续传代和体外连续传代产生HBsAg单克隆抗体(McA-HBs)的稳定性进行了研究。表明10株杂交瘤细胞在体外连续传代12周各株细胞分泌McAb的稳定性是有明显差别的,而在BALB/c小鼠体内连续传3代其产生McA-HBs的能力均较稳定。 Sr-6和Sr-19在37℃和33℃两种不同培养温度下,以及在液氮中长期冻存复苏后对杂交瘤细胞产生McA-HBs的能力也无明显影响。 This paper is reporting the investigations of producing HBsAg monoclonal antibodies ( McAb ) by 10 strains of hybridoma cell lines which were subpassaged serially in vivo in BALB/c mice for 3 generations and in vitro as routing for 12 weeks. Our results showed that 9 out of the 10 tested strains were stable enough to secrete McAb in vivo. It was also demonstrated that there was no obvious effect on the ability of prodtcing HBsAg McAbs when comparing the 2 cell strains Sr-6 and Sr-19 cultivated at 37℃ and 33℃ respectively. Through a comparison of long term subpassaging between the 2 cell lines Sr-19 and Sr-19 F, no influence on the activity to secrete HBsAg McAbs was observed in relating to the long serial cultivation after recovery from freezing state in liquid nitogen.
