摘要
用鼠抗人甲状腺球蛋白单克隆抗体(18A1)免疫家兔,取抗血清流穿以正常鼠Ig与无关单抗(MIg-Lp-Sepharose 4B)制备的亲和层析柱,吸除抗同种型及同种异型抗体。经ELISA竞争抑制、中和抑制试验证明制备出了抗TG独特型抗体(TG-Ab2)。用此TG-Ab2包被固相载体,建立了两种竞争ELISA法检测针对不同抗原决定簇的抗TG单抗之间的CRI。实验结果显示,抗TG单抗间存在CRI,从而提示自身抗体的产生可能是受胚系基因所控制。
Anti idiotypic antisera were prepared in rabbits immunized with monoclonal anti-thyroglobulin antibody ( 18A1 ). The obtained antisera with anti-Id specificily were made by a series of adsorption on Sepharose 4B immunoadsorbents coated with normal mouse immunoglobulin ( MIg ) and anti-legionclla pneumophila McAb(Lp-1-32 IgG1 ) , which shares isotype and allotype with the McAb. The results of competitive inhibition ELISA and binding inhibition EL1SA indicated ihai activity of antithyroglobulin anti-idiotypic antibody ( TG-Ab2 ) was obtained. Idiotypic cross reactivity of anti-thyroglobulin antibodies were tested by two methods of competitive ELISA. The results showed that there were CRI among murine monoclonal antibodies to thyroglobulin. It suggested that the production of autoantibody may be restricted to germline genes.
