摘要
目的研究人参皂苷Rh2对肝癌细胞表皮生长因子受体(EGFR)表达的影响,以期为人参皂苷Rh2的抗肿瘤机制提供实验证据。方法采用实时定量荧光PCR、Western blot、MTT法、荷瘤小鼠模型研究人参皂苷Rh2对肝癌细胞EGFR的表达影响,以及对肝癌细胞的生长的抑制作用。结果人参皂苷Rh2对肝癌细胞EGFR的表达具有明显的抑制作用,并且这种抑制作用具有质量浓度依赖性;人参皂苷Rh2可在体外及体内抑制肝癌细胞的增殖。结论人参皂苷Rh2抑制肝癌细胞的增殖有可能是通过EGFR通路发挥作用。
To investigate the effects of ginsenoside Rh2 on the expression of epidermal growth factor receptor (EGFR) of hepatocellular carcinoma cells, we employed the real-time quantitative fluorescent PCR and Western blot to determine the expression of EGFR mRNA and protein, and using the MTT and tumor-bearing mice model to investigate the inhibition of ginsenoside Rh2 on hepatocellular carcinoma cells proliferation. Results showed that ginsenoside Rh2 could significantly inhibit liver cancer cells EGFR expression in a concentration-dependent manner, and then inhibited the proliferation of liver cancer cell in vitro and in vivo. In conclusion, EGFR pathway may play a key role in the inhibitory effects of ginsenoside Rh2 on the proliferation of hepatoma cells.
