摘要
本文应用间接ELISA试验,混合ELISA夹心试验,ELISA抑制试验对抗B-CLL患者血清IgM同种型和独特型McAb的特异性和亲和力进行了分析。并采用ELISA双抗体相加试验,检测了McAb识别抗原的表位。试验结果表明,14株McAb中10株为抗独特型McAb,4株为抗同种型McAb。对ELISA双抗体相加试验测得结果经微机集群处理分析,可将4株抗同种型McAb分成2组,10株抗独特型McAb分成4组。ELISA相加试验结果与间接混合ELISA抑制试验结果一致。结果提示,14株McAb至少可以识别6个不同的抗原表位。
A double antibodies additivity ELISA test was employed to identify the epitopes which can be recognized by monoclonal antibodies (McAbs) against IgM from B chronic lymphocyte leukemia ( B-CLL ). The computer grouping programme analysis showed that 4 anti-isotypic McAbs could be divided into two groups and 10 anti-idiot-ype McAbs could be divided into four groups. The result was consistent with that of the indirect sandwich ELISA and inhibition ELISA test. It suggested that there were at least 6 distinct IgM epitopes which can react specifically with 14 McAbs. Our study indicated that the combination of the additivity ELISA test and the computer grouping programme analysis is of help in studying the relationship of the structure and function of antigen.
