摘要
目的探讨氧化型低密度脂蛋白(ox-LDL)及荷叶生物碱对THP-1源性巨噬细胞中血凝素样氧化型低密度脂蛋白受体1(LOX-1)表达的影响。方法①用不同浓度(0、25、50、100mg/L)ox-LDL刺激人单核细胞系THP-1源性巨噬细胞;②预先用不同浓度(0、25、50、100mg/L)荷叶生物碱处理,再予50mg/Lox-LDL培养;荧光实时定量PCR和Westernblot检测各组细胞LOX-1mRNA和蛋白的表达。结果 ox-LDL各剂量组皆可上调LOX-1mRNA及蛋白水平,在0~50mg/L剂量范围内呈明显的剂量-效应关系(P<0.01)。荷叶生物碱处理组细胞LOX-1mRNA及蛋白水平呈浓度依赖性降低(P<0.01)。结论 ox-LDL可上调THP-1源性巨噬细胞LOX-1的表达;荷叶生物碱可抑制ox-LDL诱导的THP-1源性巨噬细胞LOX-1的表达。
Objective To investigate the inhibitory effects of nuciferine on the expression of Iectin-like oxidized low- density lipoprotein receptor 1 (LOX-1) induced by oxidized low density lipoprotein (ox-LDL) in THP-1 derived mac- rophages. Methods ①THP-1 derived macrophages were stimulated by different concentrations of ox-LDL (0, 25, 50, 100 mg/L,respectively) ;②THP-1 derived macrophages pretreated with different concentration of nuciferine (0, 25, 50,100 mg/L, respectively) were incubated with ox-LDL (50 mg/L);The mRNA and protein expression of LOX-1 were determined by Sybr Green reverse transcription-polymerase chain reaction (PCR) and Western blot. Results mRNA and protein expression of LOX-1 were upregulated by ox-LDL in a concentration-dependent manner within 0-50 mg/L(P〈0.01). The stimulated effects were inhibited by pretreating with nuciferine in a concentration-dependent manner (P〈0.01). Conclusions Nuciferine showed an inhibitory effect on the expression of LOX- 1 induced by ox-LDL in THP-1 derived macrophages.
