过氧化物酶6与精子活力相关性的研究 被引量:5

摘要 目的探讨过氧化物酶6(PRDX6)与精子活力的相关性,以及PRDX6与热休克蛋白70-2(HSPA2)的关系。方法 107例精液标本,按照精子活力分组,A组(a+b)≤20%,B组为30%≤(a+b)<40%,C组(a+b)≥40%。提取精子蛋白,经聚丙烯酰胺凝胶电泳(SDS-PAGE)分离后,Western-blot检测精子质膜PRDX6的表达情况;通过凝胶成像分析系统分析精子中PRDX6和HSPA2的IOD值,用统计学软件分析两者相关性及与活力的关系。结果 A、B、C三组精子标本,质膜PRDX6表达率分别为81.5%、70.0%和17.1%,A组PRDX6阳性检出率显著高于其他两组(P<0.01);PR DX6与HSPA2的表达量呈正相关(r=0.713,P=0.000),两者分别与精子活力相关。结论精子膜PRDX6表达与精子活力相关,提示其可能是精子活力的指标;精子全蛋白中PRDX6与HSPA2相关,提示其与精子成熟度相关。 Objective: To investigate the correlation of peroxiredoxin 6 (PRDX6)and sperm motility & sperm maturity. Methods. One hundred and seven semen samples were divided into three groups according to sperm motility:group A,(a + b)≤ 20%,group B,30% ≤(a + b)〈40% and group C,(a + b)≥ 40%. The sperm proteins were extracted and run SDS-PAGE gel. The expression of PRDX6 in sperm plasma membrane was detected by Western blot. The IOD values of PRDX6 and heat shock protein 70-2(HSPA2) were determined by gel imaging analyzer. The correlation between PRDX6 and HSPA2 and the relationship between the expression of PRDX6 & HSPA2 and sperm motility were analyzed. Results: The positive rate of plasma membrane PRDX6 in group A, B and C was 81.5 %,70.0% and 17.1% respectively. The percentage in low motility group was much higher than that in high motility group. The expression of PRDX6 and HSPA2 in sperm was positively correlated sperm motility. Conclusions: The expression of PRDX6 in sperm plasma membrane is positively correlated with sperm motility,which suggests PRDX6 may be a marker of sperm motility. The expression of PRDX6 is also correlated with HPSA2 in sperm,which suggests that PRDX6 is related with sperm maturity as HPSA2 is an indicator for sperm maturity.
