摘要
用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)比色法筛选乳源性多肽,进一步采用体外模拟胃肠道消化的方法,检测经过胃蛋白酶和胰酶酶解后的合成多肽对小鼠脾脏淋巴细胞体外增殖的影响,并采用超高效液相色谱-四极杆飞行时间质谱(U-PLC-Q-TOF-MS)分析模拟消化后的产物。结果表明,QEVPL具有较好的刺激淋巴细胞增殖能力,经体外模拟消化,终产物促进淋巴细胞增殖的能力下降至原来的93.71%,但没有显著差异。质谱分析结果显示,QEPVL体外模拟消化的终产物为QEPV。进一步的验证实验表明,消化产物QEPV的刺激指数为1.1466,仍具有促进淋巴细胞增殖能力。
In this study, we evaluated the immunological activity of several milk-derived peptides by MTT assay. And the most active one QEPVL was subjected to pepsin and Corolase PP to simulated gastrointestinal digestion in vitro. The results show that there is no significant change of the immunological activity before and after digestion. With the UPLC-Q-TOF-MS, we identified the hydrolysates and found that QEPVL was hydrolyzed into QEPV. For the further confirmation, we found that the SI of QEPV was 1.466, which coincide with our former experiment.
