摘要
建立一种用4,5-亚甲基二氧基-1,2-邻苯二胺盐(DMB)衍生的荧光高效液相色谱(HPLC-FLD)检测婴儿配方奶粉中唾液酸的方法,该方法首先酸水解,然后避光衍生后荧光高效液相色谱测定。用c18柱分离,流动相为甲醇∶乙腈∶超纯水=7∶8∶85,流速为1 mL/min,标准曲线法定量。结果表明:唾液酸在甲酸水解作用中从糖链、糖蛋白中释放出来,达到了最大的释放量;并在50℃,150 min条件下DMB避光衍生,达到反应的最大值,并且该方法具有良好的准确度和精密度,平均回收率为95.83%:,最低检出限为0.02μmol/L。
