菲律宾蛤仔寡肽体外抗氧化、抗肿瘤活性研究

Studies in vitro on the Antioxidant and Anticancer Activity of Oligopeptide Isolated from Ruditapes philippinarum Hydrolysate

摘要 目的:研究菲律宾蛤仔酶解寡肽的体外抗氧化、抗肿瘤活性。方法:采用分光光度法测定寡肽对2,2'-连氮基-双-(3-乙基苯并二氢噻唑啉-6-磺酸)二铵盐(ABTS)、超氧阴离子(O2-.),羟自由基(.OH)的清除能力和寡肽的金属离子螯合能力,及四甲基偶氮唑蓝(MTT)法检测寡肽对肿瘤细胞DU-145的增殖抑制效应。结果:菲律宾蛤仔酶解寡肽有较显著的ABTS自由基[半数有效浓度(EC50)0.204 g.L-1]、羟自由基(EC500.815 g.L-1)和超氧阴离子自由基(EC500.066 g.L-1)清除能力和金属离子螯合能力(EC501.554 g.L-1)及有效的DU-145细胞增殖抑制活性(半数抑制浓度(IC50)5.202 g.L-1)。结论:菲律宾蛤仔酶解寡肽具有较好的抗氧化及抗肿瘤活性,可能与其低相对分子质量及蛋白酶酶解构成寡肽的结构相关。 Objective: To study the antioxidant activity of oligopeptide isolated from Short-neck clam (Ruditapes philippinarum) muscle hydrolysate. Method: The antioxidant activities were evaluated by using 2,2'-azinobis -diammonium salt (ABTS), hydroxyl, and superoxide anion radical scavenging assay, and Fe2+ chelation assay. The anti-prostatic cancer activity was evaluated by MTT assay. Result: Ruditapes philippinarum oligopeptide exhibited potent scavenging activity on ABTS radical (EC50 0.204 g·L^-1), superoxide anion (EC50 0.066 g·L^-1), and hydroxyl radical (EC50 0.815 g·L^-1), and significient Fe2+ chelation (EC50 1.554 g·L^-1) ability and proliferation inhibition ability (IC50 5.202 g·L^-1) of DU-145 cells. Conclusion: The antioxidant and anti-prostatic cancer activities of oligopeptide were due to the small molecular weight and special oligopeptide sequence.
