基于体外抗氧化能力探讨枳葛橘对酒精性肝氧化损伤的保护作用 被引量:1

In vitro Antioxidant Ability to Evaluate Compatibility Ratio of Formulation Zhigeju on Protection in Alcoholic Liver Injury

摘要 目的:以体外抗氧化能力初步筛选复方枳葛橘对酒精性肝氧化损伤保护作用的最佳配伍比。方法:采用正交试验法设计枳椇子、葛花和化橘红的不同配伍比,考察不同配伍比处方水提液清除1,1-二苯基-2-三硝基苯肼[1,1-Diphenyl-2-picrylhydrazyl radical 2,2-diphenyl-1-(2,4,6-trinitro phenyl)hydrazyl,DPPH.],羟自由基(hydroxyl radical,.OH)和超氧阴离子(superoxide anion free radical,O2-.)等的能力,综合评价其体外抗氧化能力,以抗氧化能力强者为复方枳葛橘的最佳配伍。结果:正交试验分析得到复方枳葛橘对酒精性肝氧化损伤保护作用的最佳配伍比例为枳椇子-葛花-化橘红(3∶3∶2),在此配伍比例下复方水提液的综合抗氧化能力较强,在生药质量浓度1.0 g.L-1下对DPPH,O2-.和.OH的清除率分别为69.94%,32.47%,58.81%。结论:复方枳葛橘体外抗氧化最佳配伍比为枳椇子-葛花-化橘红(3∶3∶2),在此配伍比条件下其综合抗氧化能力较强。 Objective: To select the compatibility ratio of formulation Zhigeju water extracts which have the best protective effect on alcoholic liver injury by in vitro antioxidant activity. Method: The orthogonal test method was employed to design the different compatibility ratio of Hovenia Dulcis-Flos Puerar and exocarpium Citrus Grandis. The antioxidant assays (DPPH, O^2-·,·OH scavenging activity) were employed to evaluate the antioxidant activities of the water extracts, and select the optimal compatibility ratio of formulation Zhigeju by the best total antioxidant ability. Result: The orthogonal test analysis got the best compatibility ratio was Hovenia Dulcis-Flos Puerar-exocarpium Citrus Grandis 3: 3: 2, the total antioxidant ability of water extraction was strongest at the ratio. The clearance rate of DPPH·, superoxide and hydroxyl free radical were 69.94%, 32.47%, 58.81% at the concentration of crude drugs 1.0 g·L^-1. Conclusion: Formulation Zhigeju's best compatibility ratio of antioxidant ability is Hovenia Dulcis-Flos Puerar-exocarpium Citrus Grandis 3: 3: 2, and their total antioxidant abilities were better.
