摘要
目的 探讨子宫内膜异位症患者在位子宫内膜表面主要组织相容性复合物 Ⅰ类 (majorhistocompatibilitycomplex classⅠ ,MHC Ⅰ )抗原的表达及其调控。方法 分离 9例子宫内膜异位症患者在位子宫内膜间质细胞 ,采用流式细胞仪 (flowcytometry ,FCM)检测其表面MHC Ⅰ抗原水平 ;经体外培养和γ 干扰素调节间质细胞表面MHC Ⅰ抗原水平 ,并测定不同MHC Ⅰ抗原水平的内膜间质细胞对淋巴因子激活杀伤 (lymphokine activatedkiller,LAK)细胞活性。结果 间质细胞表面MHC Ⅰ抗原的基础表达有变异 ,但幅度不大 ,FCM荧光强度主要分布于 10 1~ 10 2 之间 ;体外培养 2周后 ,间质细胞MHC Ⅰ抗原水平明显降低 ,FCM荧光强度分布明显左移 ,主要分布于 10 0 ~ 10 1之间 ,对LAK细胞毒作用敏感 (LAK活性为 5 1 2 8% ) ;而体外培养后再经γ 干扰素处理过的间质细胞MHC Ⅰ抗原恢复至新分离水平 ,对LAK细胞毒作用产生抵抗 (LAK活性为 2 8 6 4% )。结论 子宫内膜间质细胞上MHC Ⅰ抗原对自然杀伤 (NK)和NK样T细胞毒性具有抑制作用 ,而间质细胞表面MHC
Objective To study the possible effect of major histocompatibility complex class Ⅰ (MHC Ⅰ) antigen on endometrial stromal cells in endometriosis patients. Methods The stromal cells of endometrium was separated and cultured in vitro. γ interferon (γ IFN) was added to modulate the MHC Ⅰ antigen level which was assessed by flowcytometry, and the sensitivity of endometrial stromal cells to cytotoxic lymphocytes was examined by methyl thiazolyl tetrazolium (MTT). Results Our study demonstrated that in vitro cultured stromal cells with significant reduced MHC Ⅰ antigen levels showed enhanced susceptibility to lysis (51.28%) mediated by lymphokine activated killer (LAK) cells, whereas γ IFN treated in vitro cultured stromal cells with restored MHC Ⅰ antigen levels showed more resistance to lysis (28.64%). Conclusions MHC Ⅰ antigen on endometrial stromal cells may inhibit the cytotoxicity of natural killer (NK) cells and NK like T cells, which might play a key role in the pathogenesis of endometriosis. The level of MHC Ⅰ antigen could be regulated by culture or some biologic factors such as γ IFN.
