Effect of Electroacupunture in Different Frequency on Matrix Metalloproteinase-9 in Asthmatic Guinea Pigs with Airway Remodeling

摘要 目的:观察不同频率电针对哮喘豚鼠气道重构模型基质金属蛋白酶-9(MMP-9)表达的影响。方法:健康豚鼠32只,随机分为4组,正常对照组、模型组、低频组和高频组,每组8只。用100g/L卵清蛋白注射液进行腹腔注射以致敏,从第15天起,隔天用10g/L卵蛋白溶液超声雾化以激发(共6周)进行造模。分别采用低频和高频两种不同频率的电针针刺背三针(大杼、风门、肺俞),共28天。检测各组支气管和肺组织中MMP-9的表达。结果:模型组豚鼠支气管肺泡组织中MMP-9蛋白表达高于正常对照组(P<0.01),提示造模成功。经过电针治疗后豚鼠支气管肺泡组织中MMP-9蛋白表达降低,低频组、高频组与模型组比较,差异均有非常显著性意义(P<0.01)。低频组效果优于高频组(P<0.01),并且低频组MMP-9蛋白表达与正常对照组接近(P>0.05)。结论:电针豚鼠背三针可通过下调MMP-9表达,以干预哮喘豚鼠的气道重构,且低频电针优于高频电针。 Objective: To observe the effect of electroacupuncture (EA) in different frequency on matrix metalloproteinase-9(MMP-9) in asthmatic guinea pigs with airway remodeling. Methods: Thirty-two healthy guinea pigs were equally randomized into 4 groups, normal control group, model group, low-frequency EA group and highfrequency EA group. The guinea pig model was established by sensitization with intraperitoneal injection of 100g/L ovalbumin(OVA) on the first day, and by challenge with nebulized inhalation of 10g/L OVA solution on the 15th day, every other day for 6 weeks. Every time before challenge, the interventive treatment of high and low-frequency EA on back six points(Dahu, Fengmen, Feishu) was carried out. After treatment for 28 days, the expression of MMP-9 in the bronchi and lung was detected by immunohistochemical method. Results: MMP-9 expression in the bronchovesicular of model group was obvious as compared with that in the normal control group(P〈0.01), indicating the success of the modeling. EA decreased the expression of MMP-9 of model rats(P〈0.01 compared with the model group), and the effect of low-frequency EA was superior to that of high-frequency EA(P〈0.01). The MMP-9 expression level in low-frequency EA group was similar to that in the normal control group(P〉 0.05). Conclusion: The mechanism of EA of back six points on counteracting airway remodeling in asthmatic guinea pigs is probably related with the down-regulation of MMP-9 expression, and low-frequency EA shows better effect than hi qh-frequencv EA.
