rHSG基因对COPD大鼠气道成纤维细胞增殖凋亡的调控 被引量:4

摘要 目的:研究外源性大鼠增殖抑制基因(rat hyperplasia suppressor gene,rHSG)对慢性阻塞性肺疾病(COPD)大鼠气道成纤维细胞增殖的影响。方法 :采用阳离子脂质体将重组质粒pEGFP-rHSG转染至COPD模型大鼠气道成纤维细胞,分别感染24、48 h及3、5 d和7 d后,用实时荧光定量PCR(Real time-PCR)检测rHSGmRNA的不同时段表达水平,MTT、流式细胞仪检测转染后成纤维细胞增殖及凋亡。结果:转染24 h,rHSGmRNA表达量开始增加,高峰在7 d时,各转染组间rHSG mRNA表达差别满足(P<0.05);转染48 h后rHSG基因转染组成纤维细胞的增殖抑制明显低于未转染组(P<0.05);转染后48 h凋亡开始,与对照组相比,随着时间延长,凋亡逐渐增加,7 d时凋亡率达最大(P<0.05)。结论:外源性rHSG基因能够抑制气道成纤维细胞增殖诱导其凋亡。 Objective To study the effects of exogenous rat hyperplasia suppressor gene(rHSG) on the proliferation of airway fibroblasts of rats with COPD.Methods The recombinant plasmid pEGFP-rHSG was transfected into airway fibroblasts of rats with COPD,then the expressions of rHSG mRNA at 24 h,48 h,3 d,5 d,7 d after transfection were detected by real-time fluorescence quantitative polymerase chain reaction(real timePCR),and the proliferation and apoptosis of the transfected airway fibroblasts were measured by MTT and flow cytometry.Results The expression of rHSG mRNA began to increase at 24 h after transfection,and reached peak at 7 d.The proliferation of airway fibroblasts was significantly inhibited in rHSG transfection group than that in model control group at 48 h after transfection(P 0.05),while the apoptosis rate of airway fibroblasts was increased at 48 h,and reached peak at 7 d after transfection(P 0.05).Conclusion Exogenous rHSG can suppress the proliferation and induce apoptosis of airway fibroblast.
