转录因子8在食管癌细胞EC1的表达及其对EC1细胞迁移侵袭的影响

摘要 目的:研究食管癌细胞EC1中转录因子8(transcription factor 8,TCF8)的表达对EC1细胞迁移侵袭的影响。方法:通过共聚焦显微镜观察EC1细胞中TCF8的表达;吡咯烷二硫代氨基甲酸盐(pyrrolidinedithiocarbamate,PDTC)作用于EC1细胞24 h后,Western blot法检测NF-κBp65、TCF8和E-cadherin蛋白表达变化,划痕损伤和transwell实验检测对细胞转移和侵袭的影响。结果:TCF8表达于EC1细胞核,PDTC可以导致EC1细胞中NF-κBp65、TCF8表达降低,而E-cadherin表达增高;PDTC组细胞迁移距离和穿过的细胞数明显低于对照组。结论:食管癌细胞EC1中表达TCF8,下调TCF8可上调E-cadherin,抑制EC1细胞的迁移侵袭,为阐明食管癌侵袭转移的分子机制提供理论依据,为临床治疗提供分子靶点。 Objective To investigate the expression of transcription factor 8(TCF8) in the esophageal carcinoma EC1 cell and to analyse its effects on the migration and invasion of the esophageal carcinoma.Methods Observe the expression of TCF8 on EC1 cell by laser scanning confocal microscope.Then,pyrrolidine dithiocarbamate(PDTC) was used to the experimental group for 24 h,the expression of NF-κBp65、TCF8 and Ecadherin was detected by Western blotting.Cell migration and invasion was measured by healing and Transwell assay.Result The expression of TCF8 was at the nucleus.After using PDTC for 24h,the expression of NF-κBp65 and TCF8 were decreased and E-cadherin was increased.The number of migrative and invasive cells in PDTC group was lower than control group.Conclusion TCF8 is expressed in EC1 cell.Decreasing of TCF8 can increase E-cadherin and inhibit migration and invasion of EC1 cells.It is beneficial to elucidate the mechanism of migration and to find a molecule taget in clinical treatment in esophageal cancer.
