反复食管内灌注盐酸诱导豚鼠食管支气管反射发生可塑性改变 被引量:4

摘要 目的:本研究旨在观察慢性食管内灌注盐酸是否可以诱导豚鼠迷走神经相关的食管-支气管反射通路神经肽表达发生改变。方法:18只豚鼠,体重(400±50)g,随机分为3组:空白对照组,NS组和盐酸组。将胃管插入豚鼠食管中、下端,灌注含0.5%胃蛋白酶的0.1 mol/L HCl,连续14 d。灌流固定,取气管、支气管、肺组织、结状神经节,免疫组化法检测SP、NK-1、NKA、NKB的表达程度及表达部位变化,结果用半定量方法表示(灰度)。结果:盐酸组气道黏膜、结状神经节SP、NKA表达明显增强,显著高于空白组和NS组(P<0.05及P<0.01)。结论:慢性食管内灌注盐酸可以诱导豚鼠产生气道神经源性炎症,及迷走神经介导的食管-支气管反射通路发生神经化学改变,提示胃食管反流相关的呼吸疾病中有神经中枢的参与。 Objective To investigate if repeated intra-esophageal instillation of hydrochloric acid(HCl) may induce the change of neuropeptide in esophageal-bronchial reflex pathway associated with vagus nerve in guinea pig.Methods 18 guinea pigs,weighted(400 ± 50)g,were randomized into 3 groups: bland control group,normal saline group(NS group),and HCl group.Animals in HCl group received instillation of 0.1 mol/L of HCl containing 0.5% pepsin at middle and lower part of esophagus once a day for 14 days.The expressions of substance P(SP),neurokinin 1(NK1),neurokonin A(NKA),and neurokinin B(NKB) in the airway and nodose ganglion were detected by immunohistochemistry,and were assessed semi-quantitatively.Results The expressions of SP and NKA in airway and nodose ganglion in HCl group were significantly higher than those in bland control group and NS group(P0.05 and P0.01).Conclusion Repeated intra-esophageal instillation of HCl can induce airway neurogenic inflammation and neurochemical alternations in esophageal-bronchial reflex pathway mediated by vagus nerve in guinea pigs,which indicated that nerve ganglia were involved in gastroesophageal reflux associated respiratory diseases.
