摘要
本试验主要对减蛋综合征病毒河北分离株(EDSV-HB)的六邻体(Hexon)基因进行了扩增、克隆、测序、表达及免疫原性的试验。将EDSV-HB株六邻体基因克隆至pET-32a-c(+)原核表达载体进行表达。重组蛋白免疫6周龄鸡,间接ELISA方法检测产生的抗体水平。结果显示,表达的重组蛋白能刺激机体产生较高抗体水平。由此表明,体外表达的六邻体蛋白能够保留天然蛋白具有的抗原性。
A strain of EDSV was detected from chickens which suffered from egg drop syndrome (EDS) in HeBei province and named as HeBei strain(EDSV-HB). The cloned genomic DNA was sub-cloned into prokaryotic expression vector PET-32a-(+). After the conditions were optimized, the recom-binant protein was expressed on a larger scale. Chickens were immuned with purified recombinant protein, and all the chickens produced relatively higher level antibodies. The results showed that the recombinant protein expressed in E. coli still maintained some antigenicity of EDSV antigenes. It provided the basis for further study on recombinant vaccine against EDS.
